Characterizing and correcting camera noise in back-illuminated sCMOS cameras

With promising properties of fast imaging speed, large field-of-view, relative low cost and many others, back-illuminated sCMOS cameras have been receiving intensive attention for light level in the past several years. However, due to pixel-to-pixel difference camera noise (called non-uniformity) cameras, researchers may hesitate use them some application fields, sometimes wonder whether they should optimize non-uniformity their before using a specific scenario. In this paper, we systematically characterize impact different types on image quality perform corrections these three representative algorithms (PURE, NCS MLE ). We verify that it is possible appropriate correction methods push major noise, including readout offset, photon response, satisfactory conventional microscopy single molecule localization microscopy. further find out that, after corrections, global read becomes concern limits performance cameras. believe study provides new insights into understanding also useful information future development technology.
